That's the most difficult part. There are many possibilities :

- With the feeling or with a stopwatch wich helps you to calculate the right time.

- Open your .srt subtitle, look at the second line and note that time :
00:00:06,259 --> 00:00:09,161
Now play your movie and stop it when the first sentence is gonna be said by the actor. Now calculate the difference between the time displayed by your video player and the time you have noted from the subtitle file.

- First sync your subtitle and your movie with your video player. Then note the time that you have needed to synchronize your subtitle and report it into Move Your Sub.
